The eukaryotic microsomal P450 system contains two components:
NADPH:P450 reductase (CPR), a flavoprotein containing both FAD and FMN,
and P450. CPR appears to be a fusion protein consisting of domains which are
homologous to flavodoxin
(FMN domain) and
ferredoxin:NADP+ reductase (FAD domain).
A soluble monooxygenase P450BM-3 from Bacillus megaterium
exists as a single polypeptide chain with two functional parts, the haem (P450)
and flavin (CPR) domains, and represents a unique bacterial
one-component P450 system. Mammalian nitric oxide synthase (NOS)
contains FAD, FMN and haem and was the first eukaryotic
self-sufficient P450-like system described. The C-terminal domain
of NOS is clearly homologous to CPR, whereas its N-terminal (oxygenase)
domain has a unique fold different from that of P450s.
